Abstract Background Inflammatory bowel disease is a chronic and excessive inflammation of the colon small intestine. We previously reported that priming mesenchymal stromal cells (MSCs) with poly(I:C) induced them to express indoleamine 2,3-dioxygenase (IDO). tried find out whether IFN-γ poly(I:C)-primed MSCs have better therapeutic efficacy on experimental colitis in IDO1-dependent manner. Methods To compare effects between unstimulated primed murine colitis, mice (C57BL6) were administered 2.5% dextran sodium sulfate (DSS) drinking water for 5 days injected intraperitoneally 1 3 following DSS ingestion. The activity index score body weight loss assessed daily until day 9. Results Mice receiving showed reduced less loss. Colon tissue from same presented attenuated pathological damage, increased Paneth cells, IDO1-expressing proliferation enterocytes. MSC treatment upregulated mRNA expression intestinal stem cell markers ( Lgr5 , Olfm4 Bmi1 ), enterocyte differentiation Muc2 Alpi Chga occludin regulatory T (Treg) Foxp3 ). decreased inflammatory infiltration lymphoid organs level pro-inflammatory cytokines IL-1β TNF-α IL-6 MCP-1 ) tissue. Notably, vivo pharmacologic inhibition IDO1 blocked upregulation diminished protective MSC. Conclusions promising new strategy improve worth further research.
